Differential microglial inflammatory responses play a role in the establishment of differentiation and maturation of oligodendrocytes in brain white matter. In a murine TBI model induced by controlled cortical action, we investigated the role of a Na+/H+ exchanger, an essential microglial pH regulatory protein, in microglial proliferation and differentiation. In the Cx3cr1-CreERT2 control mice and selective microglial Nhe1 knockout mice, similar TBI-induced contusion volumes were found. The Nhe1 cKO mice, compared to the Ctrl mice, displayed increased resistance to initial TBI-induced white matter damage. These findings collectively showed that blocking NHE1 protein promotes restorative microglial growth and oligodendrogenesis, which contributes to faster white matter repair and neurological function recovery after TBI. The effect of Nhe1 deletion in microglia on post-TBI white matter repair and neurological function recovery is unclear. Overall plan: The investigation of the effects of the deletion of Nhe1 in microglia on post-TBI white matter repair and neurological function recovery is based on a scientific research.
