Here we invented a repeat-specific hybridization chain reaction as an alternative method for the detection of repeat RNA foci in two neurodegenerative disorders: C9orf72-associated ALS and frontotemporal dementia, as well as Fragile X-associated tremor/ataxia syndrome. We found that integrated stress response activation significantly increased the number of endogenous G4C2 repeat RNA foci and triggered their selective nuclear accumulation in patient fibroblasts and patient derived neurons using R-HCR. R-HCR can be a useful tool for monitoring repeat expansion mRNA in C9 ALS/FTD and other repeat expansion disorders, according to these results.

Molecular Mechanisms for CFIm-Mediated Regulation of mRNA Alternative Polyadenylation.

In higher eukaryotes, alternative mRNA processing is a necessary step for proteome expansion and gene regulation. Here we show that CFIM functions as an enhancer-dependent initiator of mRNA 3' processing. The surprising functional similarities between CFIm and SR proteins suggest that interactions between RS-like domains of regulatory and primary factors may lead to a common activation mechanism for mRNA 3' processing, splicing, and potentially other steps in RNA metabolism.
