PPAR-γ Agonist Pioglitazone Restored Mouse Liver mRNA Expression of Clock Genes and Inflammation-Related Genes Disrupted by Reversed Feeding

"This research investigated the effect of PPAR-u03b3 agonist pioglitazone on the mRNA expression profile of principle clock genes and inflammation-related genes in the mouse liver, which could be affected by switching diet. " Mice in daytime feeding groups received food from 7 AM to 7 p. m. , and mice in nighttime-feeding groups were fed from 7 p. to 7 a m. The null hypothesis was that PG and the time of its administration had no influence on the clock gene expression, which was reduced by reverse feeding. An increase in Per2 and Rev-erb alpha mRNA at noon, an increase in Ppar-u03b3 mRNA at midnight, and a decrease in Nfu3bab mRNA at noon were among the daytime-feeding group's mRNA at noon. In general, the PG administration at 7 p. m. slightly normalized the decreased expression of clock genes and raised anti-inflammatory potency in patients who were delayed by reversed feeding. The decrease in NF-u03baB resulted in serum ceruloplasmin and IL-6 in liver tissue, as well as IL-6. According to our results, PG intake at 7 p. m. corresponds to a significant rise in Nrf2 and, in particular, Ppar-u03b3 and PPAR-u03b3 expression in daytime-feeding mice with a significant decrease in clock gene expression.
