Clinical Implications of Krüpple-like Transcription Factor KLF-14 and Certain Micro-RNA (miR-27a, miR-196a2, miR-423) Gene Variations as a Risk Factor in the Genetic Predisposition to PCOS

This research has characterized the biochemical and endocrine characteristics in PCOS patients with a comprehensive serum test, as well as the effect of allelic variations for miRNAs 27a, 196a2, 423, and transcription factor KLF14 gene polymorphism on PCOS risk and susceptibility to PCOS. Both biochemical and endocrine biomarkers that were analyzed in the study showed statistically significant changes in PCOS patients, including lipids, T2DM markers, and hormones. The study found that in the codominant model, KLF14-AA was strongly associated with increased PCOS susceptibility, miR-27a-GA was linked to an elevated PCOS susceptibility, and miR-196a-CT was associated with higher PCOS susceptibility. Moreover, allele A of KLF-14 and allele T of miR-196a2 were strongly associated with PCOS susceptibility in the target population.

Micro-RNA expression in cisplatin resistant germ cell tumor cell lines

Background: In order to expand our knowledge of cisplatin resistance mechanisms, we compared microRNA expression in three cisplatin resistant sublines derived from paternal cisplatin sensitive germ cell tumor cell tumor cell lines. Methods Three cisplatin resistant germ cell tumor cell lines, three well established relatively cisplatin sensitive germ cell tumor cell lines, three cisplatin resistant sublines, which resulted in an increase in drug resistance after intermittent exposure to increasing doses of cisplatin showing 2. 7-11. 3 fold rise in drug resistance after intermittent exposure to increasing doses of cisplatin. Cells were cultured and total RNA was isolated from all six cell lines in three independent experiments. The number of genes showing differential expression in more than one of the cell line pairs was 34 between NTERA-2R/NTERA-2 and NCCIT-R/NCCIT, respectively, and 3 and 4, respectively, between these two cell lines and 2102EP/2102EP. In all three cell lines, only the has-miR-10b involved in breast cancer invasion and metastasis, as well as has-miR-512-3p appeared to be up-regulated.

Micro RNA‐4651 Serves as a Potential Biomarker for Prognosis When Selecting Hepatocellular Carcinoma Patients for Postoperative Adjuvant Transarterial Chemoembolization Therapy

MicroRNA 4651 was used to determine the effects of paTACE therapy, according to a hospital-based retrospective study, which included 302 patients with advancedstage hepatocellular carcinoma who underwent tumor resection or tumor resection as an initial treatment. The sensitivity of tumor cells to doxorubicin was determined by the half-maximal inhibitory concentration. Upregulated microRNA4651 expression in tumor tissues can enhance the therapeutic response of patients with hepatocellular carcinoma on paTACE, but downregulated expression is ineffective. Conclusion: These findings indicate that increasing microRNA4651 expression may be helpful for paTACE in boosting hepatocellular carcinoma prognosis.

Serum Concentrations of Osteogenesis/Osteolysis-Related Factors and Micro-RNA Expression in ST-Elevation Myocardial Infarction

Both Atherosclerosis and bone metabolism are related to common molecular and cellular mechanisms. This research is intended to determine the serum concentration of osteogenesis/osteolysis factors panel, serum expression levels of micro-RNA-24-1 and miR-6802, and determine their relationship with myocardial injury, LV reconstruction and function in STEMI's acute phase. During the acute phase and the 3-month follow-up, there were no significant differences in serum osteogenesis/osteolysis factors and expression of miR-24-1 and miR-6102. Patients with at least 5% LVEF improvement and those without improvement were similar. During the acute phase of myocardial infarction, there was no correlation between the OPG serum level and LVEF. While the 3-month follow-up, serum osteogenesis/osteolysis factors, miR-24-1 and miR-6802 expressions in STEMI patients, as well as miR-24-1 and miR-6802 expression, do not change much in STEMI patients, and are not correlated with LV remodeling and function.

A micro RNA processing defect in rapidly progressing idiopathic pulmonary fibrosis.

The purpose of this study was to determine whether differential miRNA expression could be a factor in the development of IPF in rapid versus slowing modes. METHODOLOGY AND PRINCIPAL FINDINGS: mRNA and mRNA were isolated from surgical lung biopsies from IPF patients with a clinically confirmed fast or slow course of disease over the first year after diagnosis. The application of a quantitative PCR miRNA array containing 88 of the most abundant miRNA in the human genome was used to determine lung biopsies from 9 patients with rapidly progressing IPF, 6 patients with slowly progressing IPF, and 10 normal lung biopsies. MiRNA with validated mRNA targets were among the miRNA tested in IPF with confirmed mRNA targets, those with regulatory impacts on epithelial-mesenchymal transition. The expression of AGO1 and AGO2 in comparison to either slow or normal lung biopsies and fibroblasts grown from the same biopsies revealed by further examinations.
