Novel efficacious microRNA-30c analogs reduce apolipoprotein B secretion in human hepatoma and primary hepatocyte cells.

High blood lipid levels have been shown to raise cardiovascular disease risk. We previously reported that increasing cellular levels of microRNA-30c using viral vectors or liposomes reduces plasma lipids and atherosclerosis. We hypothesized that miR-30c's update would improve miR-30c's stability and increase liver cell delivery. Without impacting apolipoprotein B secretion and cell lipid levels, we show that these analogs have significantly reduced apolipoprotein B secretion in Huh-7 human hepatoma cells and human primary hepatocytes without effecting apolipoprotein A1 secretion and cellular lipid levels. Our findings show that the passenger strand of miR-30c can be modified to improve its consistency and delivery to cells while still maintaining the sense strand's potency.
