Abstract Background: Abstract Background: Abstract Background The aim of process development in Pichia pastoris has evolved to include a mixed feeding program in order to raise cell biomass and enzyme production rates. Results Using glucose oxidase as a model protein, the particular and synergistic effects of co-feeding of sorbitol or yeast extract with methanol and Hac1p overexpression on Gox's mystery gene expression were investigated both in shake flasks and in a laboratory fermenter. In terms of increasing protein expression and cell growth, YE is superior to sorbitol in terms of boosting protein expression and cell growth. Conclusions Co-feeding of YE by YE and codedion pathway engineering significantly improved glucose oxidase secretion, which can also be used to increase the expression of other foreign proteins in the P. pastoris system.

To determine P. pastoris origins, gene expression dynamics, and to determine global positioning of nucleosomes across the genome, we used a suite of vastly parallel sequencing techniques to map and dissect P. pastoris origins, as well as to determine the global positioning of nucleosomes around the world. Our results reveal that some functional overlap exists between promoter sequences and G/C-rich replication origins in P. pastoris and G/C-rich replication origins, implying an evolutionary bifurcation of the modes of replication origins.
