Formulation and Characterization of Stimuli-Responsive Lecithin-Based Liposome Complexes with Poly(acrylic acid)/Poly(N,N-dimethylaminoethyl methacrylate) and Pluronic® Copolymers for Controlled Drug Delivery.

Copolymers of Pluronic®-poly and Pluronic®-poly synthesized by atom transfer radical polymerization are included in the chosen hydrophilic polymeric system. The copolymers were embedded into liposomes made from soybean lecithin and with different copolymer/phospholipid ratios. Their cytotoxic activity was determined by testing the viability of human epithelial cells exposed to them. The formulated PLCs had useful characteristics, particularly the ability of stimulating rapid rises in pH and/or temperature, with no evidence of cellular involvement, which may be of concern in the treatment or diagnosis of all diseases that cause some specific pH/temperature change in the target area.

Dual stimuli-responsive dendronized prodrug derived from poly(oligo-(ethylene glycol) methacrylate)-based copolymers for enhanced anti-cancer therapeutic effect.

The introduction of the dendron in the prodrug blocked the prodrug's entanglement of brush oligo-chains, as well as gradual degradation of the polymeric prodrug's ability to produce stable nanoparticles. The double-responsive dendritic structure enhanced cell uptake and facilitated drug release in response to elevated cathepsin B and an acidic pH in the tumor microenvironment, resulting in an elevated therapeutic effect with a tumor inhibition rate of 72. 9% for 4T1 tumor-bearing mice. Hence, this option may have a great deal of insight into the manufacture of poly-based copolymers as drug delivery providers. STATEMENT OF SIGNIFICANCE: We recommend a dual-stimuli-responsive dendronized program for increasing the cancer therapeutic value of poly-based drug conjugates. The drug delivery device has a high therapeutic potentcy with negligible side effects.
