Matrix Metalloproteinases on Severe COVID-19 Lung Disease Pathogenesis: Cooperative Actions of MMP-8/MMP-2 Axis on Immune Response through HLA-G Shedding and Oxidative Stress

Patients with COVID-19 have a respiratory tract infection in most patients, and acute lung dysfunction is the most common complication. Although the clinical cause of SARS-CoV-2 immunopathology is still unknown, it is nevertheless clear that lung infection is responsible for hyper inflammation and tissue damage, as well as tissue damage. We wanted to pinpoint a situation in which MMPs could have a role in COVID-19's lung pathogenesis. We found a high number of MMPs in lung tissue from COVID-19 patients by proteomics, but we also determined the expression and enzyme activity of MMP-2 in tracheal-aspirate fluid samples from intubated COVID-19 and non-COVID-19 patients.

Prevention of interleukin-8-induced mobilization of hematopoietic progenitor cells in rhesus monkeys by inhibitory antibodies against the Metalloproteinase gelatinase B (MMP-9)

Previously, we showed that IL-8 stimulates rapid proliferation of hematopoietic progenitor cells from the bone marrow of rhesus monkeys. Because activation of neutrophils by IL-8 results in the production of extracellular matrix molecules, we hypothesized that MMP-9 release may cause stem cell mobilization by cleaving matrix molecules to which stem cells are attached. Rhesus monkeys were treated with a single i. v. rhesus monkeys were treated with a highly specific inhibitor monoclonal anti-gelatinase B antibody, in order to verify the likelihood that MMP-9 induction was involved in HPC mobilization. MMP-9 is activated by IL-8 leading to the rapid systemic release of MMP-9 with simultaneous blood mobilization of HPC that is otherwise unaffected by pretreatment with an inhibitory anti-gelatinase B antibody, indicating that MMP-9 is involved as a mediator of the IL-8-induced mobilization of HPC.

Membrane-type matrix metalloproteinase (MT-MMP) gene is expressed in stromal cells of human colon, breast, and head and neck carcinomas.

We have found MT-MMP gene expression in all 83 human carcinoma samples tested by RNA in situ hybridization and a few in tumor stroma but not in cancer cells, but not in tumor cells. The expression pattern of the MT-MMP gene was more closely related to that of the fibroblastic cells of human carcinomas than that of the stromelysin 3 or interstitial collagenase genes, according to a comparison with other MMP genes expressed in fibroblastic cells of human carcinomas.

Activated integrin αvβ3 cooperates with metalloproteinase MMP-9 in regulating migration of metastatic breast cancer cells

Metastasis in human breast cancer cells is strongly promoted by the expression of adhesion receptor integrin v3. v3 3 cooperates with matrix metalloproteinase type 9 in breast cancer cell migration, according to this article. MDA-MB 435 human breast cancer cells and primary metastatic cells from breast cancer patients have marked migration toward vitronectin and fibrinogen. Whereas all tested breast cancer cell variants produced latent MMP-9, only those with activated vs. 3 produced the mature form of this metalloproteinase. In metastatic breast cancer cells, the results show a causal connection between activated integrin vs. 3 and the production of enzymatically active MMP-9. These molecules work together to promote breast cancer cell migration toward specific matrix proteins, and this can lead to a significantly elevated metastatic capacity of breast cancer cells that express activated vs. 3.

Metalloproteinase MT5-MMP is an essential modulator of neuro-immune interactions in thermal pain stimulation

Peripheral interactions between nociceptive fibers and mast cells contribute to inflammation pain, but no information is known about the mechanisms facilitating neuro-immune communication. MT5-MMP is expressed by CGRP-containing peptidergic nociceptors in dorsal root ganglia, and Mmp24-deficient mice have increased sensitivity to noxious thermal stimuli under basal conditions. Mmp24 / mice do not experience thermal hyperactivity during inflammation, a phenotype that is consistent with changes in N-cadherin-mediated cell-cell interactions between mast cells and sensory fibers, in comparison to basal nociception.

Association of Matrix Metalloproteinase-2 (MMP‑2) and MMP‑9 Promoter Variants, their Serum Levels, and Activities with Aortic Valve Calcification (AVC) in a Population from Western Iran

We investigated the possibility of a coordinate between MMP-2 and T alleles of MMP-9 in synergy and synergism, as well as the threat of AVC. Using PCR–RFLP, 92 cases of AVC with a median age of 61. 78 years from West of Iran were included, and 92 healthy individuals with a mean age of 61. 78 years were identified, and MMP-92G / A and MMP-9-1562 C / T promoter polymorphism was detected in 92 cases with AVC with a mean age of 61. 78 years were included, with a mean age of 62 cases with AVC In the presence of heart valve calcification disease, the frequency of A alleles from the MMP2-1575 variant was marginally higher. Both MMP-2 and MMP-9 serum levels and activity were found to be significantly higher than the control group. In patients, the MMP-2 and MMP-9 serum concentrations and activity are elevated, and the control group warns them of cardiovascular disease.

Abstract Solar ultraviolet radiation exposure to human skin is well known, and they can range from increased skin aging to skin cancer. We investigated the protective effects of HelioVital sun protection filter foil against UVA1 irradiation in skin cells. It could be shown that the HelioVital sun protection filter foil has protective properties against UVA1 irradiation-induced changes in matrix metalloproteinase expression. In addition, this report established the protective role of the HelioVital filter film against UVA1-induced ROS production and DNA damage.
