Here we use intra-LN injection of immune signal-loaded biomaterial depots to specifically monitor cancer vaccine deposition, showing how the combination of signals in and between LNs influence anti-tumor response. Vaccine therapy gives near-total immunity to the tumor fight, allowing the re-challenge 100 days later, with no additional drugs. These findings show that immunotherapies that use biomaterials for loco-regional LN therapy can produce responses that are systemic and specific, without systemically exposing patients to potent or immunotoxic drugs.
