In general, circRNA BSJs are estimated in RNA sequencing results using one of several circRNA detection methods. However, linear transcripts derived from endogenous trans-splicing can result in false-positive results both in RNA sequencing and in RT-qPCR experiments. However, linear RNAs can be degraded using an exonuclease such as ribonuclease R, so doing the RT-qPCR validation step is only necessary. Here, we used an automated workflow to administer RNase R therapy on HLF, NCIH23, and SW40 cell lines.
