Methods: Indomethacin tablets were made using three different super disintegrants; crospovidone, croscarmellose sodium, and sodium starch glycolate were produced in three different amounts by direct compression technique. In wounded gastric monolayers, we investigated whether epidermal growth factors and/or indomethacin influence cell migration, actin stress fiber production, and/or phosphorylation of FAK and tensin. During baseline and EGF-stimulated conditions, IND inhibited all three cell migration and actin stress fiber formation, increased FAK localization to focal adhesions, and increased FAK localization to focal adhesions, as well as phosphorylation of FAK and tensin. FAK phosphorylation inhibition by IND was related to shifts in actin polymerization, showing that actin depolymerization could be a result of reduced FAK activity. Pretreatment by IND significantly delayed epithelial maturation in vivo, reduced FAK phosphorylation and recruitment to adhesion points, as well as actin stress fiber formation in migrating surface epithelial cells.

Cassiopea xamachana jellyfish are an inexpensive model system for investigating metamorphosis and/or cnidarian symbiosis in the wake of their asexual cycle, in which the latter can spawn new larvae and scyphistomae without differentiation into ephyrae. Although strobilization induction and differentiation into ephyrae can be achieved in a variety of ways, no reports have been published on a controlled, reproducible metamorphosis induction. We investigated if differentiation could be pharmacologically enhanced as in Aurelia austria by the metamorphic inducers thyroxine, KI, NaI, Lugols iodine, H 2 O 2, indomethacin, or retinol. After six days of exposure, we discovered reproducibly induced strobilization by 50 % bcM indomethacin, and 10 u201325 u201327 bcM after 7 days. In short, indomethacin can be used as a quick instrument for determining biological functions by a controlled metamorphic process in C. xamachana scyphistomae.
