Biallelic variants in MOS cause large polar body in oocyte and human female infertility.

"Study question What is the underlying physiological cause of female infertility involving abnormal oocyte morphology during the development of a large first polar body? Female infertility with early embryonic arrest and fragmentation can be attributed to biallelic mutations in MOS, according to two new independent studies. However, MOS variants have not been shown to have anything to do with female infertility by the phenotype of large PB1 extrusion in human oocytes. In HEK293 cells and mouse oocytes, the pathogenic effects of the MOS protein properties and ERK1/2 activation were determined. We found three rare missense variants in MOS, including a homozygous missense form from Patient 1 in Family 1 and two compound missense variants from twin siblings in Family 2. All three patients had a high incidence of large PB1 extrusion in the oocytes. In addition, the MOS variants in comparison to wild-type MOS decreased the MOS protein content and attenuated the binding capacity with MEK1. After siMos treatment, microinjection of wild-type human MOS complementary RNAs restored a symmetric division of oocytes. With this increased knowledge, various MQS HBC methods have been outlined for both high-impedance capacitive and 50 termination cases, as well as EQS HBC for similar sized machines, over a wide distance between TX and RX. Depending on the type of applications, the final study presents the first fundamental insight into M-HBC operation and its comparison with EQS HBC, assisting HBC device designers in making informed design decisions.
