Functional genomics screening identifies aspartyl-tRNA synthetase as a novel prognostic marker and a therapeutic target for gastric cancers.

"Recent developments in high-throughput nucleic acid synthesis techniques as well as next-generation sequencing platforms have made it possible to perform functional genomics analysis using large-scale lentiviral libraries aimed at finding novel cancer therapeutic targets. " We carried out NGS-based functional genomics testing for human GC cell lines in this study using an earlier developed 6,399 shRNA library that encompasses all 2,096 human metabolism genes. Patients with elevated expression levels of DARS protein in-house tissue microarrays combined with public records revealed more advanced clinicopathological characteristics and a poor prognosis, particularly among diffuse-type GC patients with higher frequency profiles of DARS protein. Both in vitro and in vivo experiments showed that DARS inhibition resulted in dramatic growth inhibition of GC cells. DARS inhibition may also be used to other cancers, so it is warranted to investigate the expression and clinical significance of DARS in a variety of malignancies.
