In Vitro–In Vivo Correlations Based on In Vitro Dissolution of Parent Drug Diltiazem and Pharmacokinetics of Its Metabolite

After the oral administration of the parent drug, a new type of in vitro in vivo correlation is suggested in this study: The correlation of the in vitro parent drug dissolution results with the drug's metabolite data is shown in this study. From an in vitro research in 19 healthy volunteers, pharmacokinetic data for the parent drug diltiazem and its desacetyl metabolite were obtained. This type of correlation of the in vitro results of the parent compound's in vitro results with the in vivo results of the metabolite could be useful in the manufacture of drugs with active metabolites and prodrugs.

Concomitant use of rivaroxaban with non-dihydropyridine calcium channel blockers may result in an elevated incidence of systemic rivaroxaban exposure and anticoagulant activity in relation to non-DHP inhibitory enzymes and/or transporters. In rats, the aim of this research was to investigate the effects of verapamil and diltiazem on the pharmacokinetics and the prolongation of prothrombin time. With a simple linear relationship, the double-peaks in rivaroxaban plasma concentrations and the associated increase in prothrombin time are discussed in detail by a transit PK model in the absorption process.
