Below, we utilize the Proteomic Peptide Library method to produce high-resolution versions for PKA WT and PKA L205R substrate uniqueness. Making use of these versions, we created peptide substrates that show modified selectivity for specific PKA variations, and show the feasibility of discerning PKA L205R loss-of-function signaling. Through these results, we suggest that substrate rewiring may add to Cushing's syndrome disease etiology, and introduce an effective new standard for exploring mutation-induced kinase substratum rewiring in human disease.
