Hence, the anti-aging effects of chlorpropamide, glimepiride, glibenclamide, and tolbutamide were investigated, with a particular focus on chlorpropamide's interaction with mitochondrial ATP-sensitive K+ channels and mitochondrial complex II. Both MRC-5 cells and mice, Chlorpropamide delayed aging in Caenorhabditis elegans, human lung fibroblast MRC-5 cells, and reduced doxorubicin-induced senescence. Moreover, chlorpropamide's lifespan extension by chlorpropamide required complex II and elevated mtROS levels, indicating that chlorpropamide directly or indirectly affects the production of mtROS as a pro-longevity indicator.
