Establishment of a diverse head and neck squamous cancer cell bank using conditional reprogramming culture methods

Squamous cell cancer models in head and neck rely on established immortalized cell lines, which carry inherent bias due to selection and clonality. Using a u201cconditional reprogrammingu201d strategy, we established a large panel of HNSCC tumor cultures, which uses a rho kinase inhibitor and coherobe cells from a fibroblast feeder cells to promote cell survival indefinite tumor cells. Culture against patient tumor tissue DNA was established by short tandem repeat profiling. Utilizing clinical p16 staining, RT u2010PCR of HPV16/18 specific viral oncogenes E6 and E7 in RNA isolated from tumor samples, and HPV DNA sequencing, human papilloma virus testing was done. Three of the four oropharyngeal tumors were p16 and HPVu2010 positive, with maintained HPV in culture. Regardless of patient characteristics, disease site, and molecular origin, CR methods can be easily integrated to all HNSCC tumors, providing a translational research approach that properly incorporates patient and tumor diversity.
