MicroRNA-16 Represses TGF-β1-Induced Epithelial-to-Mesenchymal Transition in Human Lung Adenocarcinoma Cell Line.

Background The transforming growth factor-beta1-induced epithelial-to-mesenchymal transition has a significant effect on lung cancer cell progression and metastasis. Objective The aim of this research was to see if microRNA -16 would reduce TGF-induced TGF-u03b21-induced EMT and cell proliferation in a human lung adenocarcinoma cell line. TGF-u03b21 in A549 cells was found to have significantly reduced miR-16 expression. Moreover, the agomir of miR-16 also reduced TGF-u03b21-induced EMT and cell proliferation, which was also inhibited by miR-16. In addition, a miR-16 mimic was found to reduce TGF-u03b21-induced activation of the TGF-Smad3 pathway, showing that miR-16 may play a part in controlling Smad3. Conclusions In A549 cells, we found that overexpression of miR-16 suppressed Smad3 production and activation, leading to the inhibition of TGF-mediated proliferation, which ultimately halted TGF-u03b21-induced proliferation. The present findings support further investigation into miR-16's anti-cancer activity in animal lung cancer models and confirm the therapeutic potential.
