Identification of TPIT and other novel autoantigens in lymphocytic hypophysitis; immunoscreening of a pituitary cDNA library and development of immunoprecipitation assays

Physiology: Lymphocytic hypophysitis is an organ-specific autoimmune disease of the pituitary gland. Design/methods The pituitary cDNA expression library was immunoscreened for lymphocytic hypophysitis using sera from four patients with lymphocytic hypophysitis. In vitro transcription and translation immunoprecipitation assay, an influential cDNA clones from screening were tested alongside previously identified autoantigens pituitary gland-specific factor 1a and 2 assay, assay. Patients with 9/86 hypophysitis patients versus 1/90 controls had a high autoantibody reactivity against TPIT. The reactivity against TPIT was not limited to lymphocytic hypophysitis with autoantibodies, which was not present in the serum from patients with other autoimmune endocrine disorders. In comparison to 0/90 controls, 8/86 patients with lymphocytic hypophysitis had autoantibodies against any two autoantigens. Potential autoantigens with different immunoreactivity profiles in patients and controls were also identified as potential autoantigens related to vesicle processing.
