Direct control of lysosomal catabolic activity by mTORC1 through regulation of V-ATPase assembly

The amino acid sensor mechanistic target of rapamycin complex 1 in amino acid-replete settings is reduced by unknown mechanism. Here, we show that mTORC1 inhibits extracellular protein degradation by suppressing V-ATPase-mediated acidification of lysosomes. As mTORC1 production decreases, V-ATPase V_1 domains in lysosomes join membrane-integral V-ATPase V_o domains to manufacture active proton pumps. The resulting decrease in luminal pH has increased protease activity and protein degradation among the lysosomal population.
