Chemophoresis engine: A general mechanism of ATPase-driven cargo transport

However, how cargo transports are affected by chemical gradients is unclear. A container carrying reaction sites is exposed to a chemophoresis agent at the point of the elevated concentration, according to the system. Based on this, we introduce the chemophoresis engine, as a general mechanism of cargo motion that converts chemical free energy into directed movement by catalytic ATP hydrolysis. We applied the engine to plasmid movement and pattern control of ParA-ATP concentration in a ParABS system, demonstrating plasmid movement and pattern dynamics of ParA-ATP concentration in bacterial cells and vitro experiments, demonstrating plasmid equi-positioning and pole-to-pole oscillation observed in bacterial cells and vitro experiments. We also found that the chemophoresis engine can work under in vivo conditions. We'll eventually address the chemophoresis engine as one of the principal mechanisms of hydrolysis-driven intracellular transport.

FERARI and cargo adaptors coordinate cargo flow through sorting endosomes

Here we show that cargo concentration in sorting endosomes determines the length of the kiss between Rab11 and SNX1 structures, presumably by clogging the fusion stalk. The transformation of sorting endosomes from Rab11 to Rab11 structures is dependent on SNX6, while cargo retention in the Rab11 endosome is dependent on AP1. Positive endosomes from Rab11, Rab5, and Rab10 can also communicate with the SNX1 tubular network by a FERARI-dependent kiss-and-run. We recommend that FERARI, along with cargo adaptors, coordinates the vectorial movement of freight by sorting endosomes.

Here, we demonstrate that FERARI has been busy mediating the unveiling of Rab5-positive vesicles with sorting endosomes. During these encounters, cargo flows from Rab5-positive vesicles to sorting endosomes and then into Rab11-positive vesicles. The conversion of sorting endosomes into Rab11 structures is dependent on the cargo adaptor SNX6, while cargo retention in the Rab11 compartment is dependent on AP1. We recommend that FERARI, together with cargo adaptors, coordinate the vectorial movement of cargo through sorting endosomes.

The glyoxalase pathway is a common cellular metabolic pathway, with the removal of methylglyoxal a key physiological function. More specifically, new reports has shown that EVs derived from human amniotic fluid stem cells contain glycolytic pay-off phase enzymes such as glyceraldehyde-3-phosphate dehydrogenase. We wanted to find out whether MG-derived MG-H1 and glyoxalases, as well as glyoxalases, could be a novel molecule cargo in these EVs since GAPDH catalyzes the sixth step of glycolysis using a substrate GA3P, which MG spontaneously originates.

Evaluating an Inland Waterway Cargo Vessel’s Energy Efficiency Indices

Abstract: The International Maritime Organization introduced the energy efficiency standards for ships more than a decade ago, but inland navigation hasn't been affected by such laws at all. Therefore, inland waterway vessels are left with no mandatory guidelines that could push their technology into more energy efficient designs. Fortunately, there are some pioneering attempts to establish energy efficiency standards for inland vessels. In addition, a typical Danube cargo inland vessel's data is used to determine their current energy efficiency standards in accordance with provisional standards. The final results revealed discrepancies in the same vessels' energy efficiency ratings, and as a rule for inland waterway vessels, the procedure should be updated and harmonised. The report provided an insight into the current state of the Danube inland fleet's traditional configuration, as well as the emergence of energy conservation legislation.
