We modified the length of fibers in sinewaves at a small amplitude, investigated concomitant tension transients, and determined the kinetic constants of the CB cycle from the ATP and Pi effects to characterize CB kinetics. The average change in force generation step and Pi release step were not affected by the mutation, but there was a gender difference in WT mice: K 4 and K 5 were 2. 2X in males and females, rather than in females. Female MC4R/- mice relative to male MC4R -/- mice were both 1. 25x in muscles, with a constant forward rate stable and backward rate steady throughout the force generation step. In both genders, Ca 2+ sensitivity decreased marginally in mutants.
