Quantitation of Oral Cell DNA Adducts in Smokers

Use recently created high resolution mass spectrometric, high throughput DNA adductomic strategy to display for unidentified and well-known DNA adducts to identify a detailed DNA alteration trademark derived from smoking. Making use of a targeted DNA adductomic method, display for numerous DNA alterations all at once. The adducts evaluated in Specific Aim 1 will be included in a listing including endogenous, 1,3-butadiene-derived, aldehyde-derived and nitrosamine or alkylating agent-derived DNA adducts. In parallel, use DNA adductomic approaches in an untargeted mode to check out the presence of previously unidentified DNA adducts. The unidentified adducts identified by the untargeted evaluation will be consisted of in the list of targeted DNA adducts to ultimately obtain a sensitive method to examine general DNA damages arising from cigarette smoking. The listing of DNA adducts created in Specific Aim 3a will be related to investigate distinctions in the DNA damages signature in chosen examples evaluated in Specific Aim 1.
