We compared GD3 expression in tissues from patients with TSC and controls as tumors may rely on ganglioside D3 protein for mTORC1 activation and growth. We performed preclinical experiments with GD3 chimeric antigen receptor T cells as GD3 overexpression was not accompanied by apparent immune responses to the target molecule. Tsc2+u2013 tumor cells, CAR T cells significantly reduced the tumor burden in mice challenged with elevated T cell infiltration, which is correlated with increased T cell infiltration. We also treated elderly Tsc2+/u2013 heterozygous mice that produce spontaneous Tsc2+/u2013 tumors with GD3 CAR or untransduced T cells, which were then analyzed at endpoint.
