Results Here, we developed a pH-sensitive biomimetic drug delivery device based on macrophage-derived microvesicle-coated zeolitic imidazolate framework-8 nanoparticles that encapsulated the drug methotrexate. In RA, the MV/MTX@ZIF-8 nanoparticles were further modified with 1,2-distearoyl-sn-glycero-3-phosphate-N-[folate -2000] to harness the octagon-in-activated macrophages' high affinity for folic acid. MTX@ZIF-8 nanoparticles had high DLE and EE. An acidic environment could be used to produce effective drug delivery, according to an In vitro study. We also found that the activated macrophage could uptake more FPD/MV/MTX@ZIF-8 than activated cells. The best joint targeting and longest circulation time were found in the FPD/MV/MTX nanoparticles in an In vivo biodistribution experiment.
