"Abstract Background Methotrexate has been praised for Rheumatoid arthritis therapy, but, MTX does not accumulate well at inflamed sites, and long-term administration in high doses has been associated with severe side effects. Results Here, we developed a pH-sensitive biomimetic drug delivery device based on macrophage-derived microvesicle-coated zeolitic imidazolate framework-8 nanoparticles that encapsulated the drug methotrexate. In RA, the MV/MTX@ZIF-8 nanoparticles were further modified with 1,2-distearoyl-sn-glycero-3-phosphate-N-[folate -2000] to harness the octagon-in-activated macrophages' high affinity for folic acid. MTX@ZIF-8 nanoparticles had high DLE and EE. An acidic environment could be used to produce effective drug delivery, according to an In vitro study.
