"Abstract API-1, a novel small-molecule inhibitor of Akt, acts by linking to Akt and preventing membrane translocation, and has promising preclinical antitumor activity. We present a new role of API-1 in the regulation of cell FLICE-inhibitory protein levels and TRAIL-induced apoptosis in this report, independent of Akt inhibition. API-1-based API-1 activated apoptosis in several experimental cancer cell lines, including activation of caspase-8 and caspase-9. These results indicate that c-FLIP mediates development of TRAIL-induced apoptosis by API-1 has decreased, but not enough to account for API-1-induced apoptosis in API-1. "Abstract During malignant glioma due to over stimulation of growth factor receptor receptors and increased regulation of the PI3K/Akt pathway, resistance to epidermal growth factor receptor inhibitors and pro-apoptotic agents is common.
